چکیده

Glucose oxidase (GOx) has found wide range of industrial applications. Commercial GOx are produced from Aspergillus niger and selected strains of Penicllium species, however, these strains are associated with some drawbacks. Aspergillus niger produces predominantly intracellular GOx, which requires extra recovery processes thus incurs more processing cost while Penicillium sp. exhibits filamentous morphology in submerged cultivation which results in poor oxygen mass transfer and in turn affects the GOx production. Therefore, it is necessary to find an alternative strain which produces predominantly extracellular GOx and exhibits pelleted morphology in submerged cultivation. The present work deals with the isolation and identification of a new isolated GOx-producing strain, Aspergillus terreus UniMAP AA-1, which produced both the desired traits. It was found that enzymatic properties of the crude enzyme are all in-line with the typical commercial GOx properties.

Optimization of complex fermentation media for glucose oxidase production using statistical approach.

Production cost of enzyme is largely determined by the type of the strain and raw material used to propagate the strain. Hence, selection of the strain and raw materials is crucial in enzyme production. For Glucose oxidase (GOx), previous studies showed Aspergillus terreus UniMAP AA-1 offers a better alternative to the existing sources. Thus, a lower production cost could be logically anticipat...

Ultraviolet-induced mutational changes in enzyme activity of Aspergillus terreus.

In previous publications Arpai (1957) and Arpai and Valentin (1957) described the production of ultraviolet-induced mutants of Aspergillus terreus, using nondifferentiated spore material and conidia differentiated by sedimentation rate. A limited number of the irradiated strains were morphologically altered and produced more itaconic acid than did the parent strain. The products of these strain...

Thermal Characterization of Purified Glucose Oxidase from A Newly Isolated Aspergillus Niger UAF-1

An intracellular glucose oxidase was isolated from the mycelium extract of a locally isolated strain of Aspergillus niger UAF-1. The enzyme was purified to a yield of 28.43% and specific activity of 135 U mg(-1) through ammonium sulfate precipitation, anion exchange and gel filtration chromatography. The enzyme showed high affinity for D-glucose with a Km value of 2.56 mM. The enzyme exhibited ...

Expression of the Aspergillus terreus itaconic acid biosynthesis cluster in Aspergillus niger

BACKGROUND Aspergillus terreus is a natural producer of itaconic acid and is currently used to produce itaconic acid on an industrial scale. The metabolic process for itaconic acid biosynthesis is very similar to the production of citric acid in Aspergillus niger. However, a key enzyme in A. niger, cis-aconitate decarboxylase, is missing. The introduction of the A. terreus cadA gene in A. niger...

Isolation and characterization of a fungus for extracellular synthesis of small selenium nanoparticles

The use of biogenic selenium nanoparticles for various purposes is going to be an issue of considerable importance; thus, appropriate simple methods should be developed and tested for the synthesis and recovery of these nanoparticles. In this study, a fungus was isolated from a soil sample, identified as Aspergillus terreus and used for extracellular synthesis of selenium nanoparticles (Se NPs)...
